Leuprorelin
Leuprorelin is a GnRH-receptor agonist. After binding to pituitary GnRH receptors, it first causes a transient surge in luteinizing hormone (LH) and follicle-stimulating hormone (FSH) release — the so-called flare. With continuous, non-pulsatile exposure the receptors are downregulated and desensitized, suppressing gonadotropin secretion and consequently lowering sex steroids (testosterone or estradiol) to low levels. This mechanism underlies the literature-described use in hormone-dependent conditions.
Teriparatid
Teriparatide corresponds to the biologically active first 34 amino acids of human parathyroid hormone and binds the PTH-1 receptor on bone and kidney cells. The literature describes that intermittent receptor activation preferentially stimulates the activity of bone-forming osteoblasts, whereas continuously elevated PTH exposure (as in hyperparathyroidism) tends to favor bone resorption. This time-dependent difference is regarded as the mechanistic basis for the bone-anabolic effect observed in studies.
