Mechanism of action
Linaclotid
Linaclotide activates the guanylate cyclase-C receptor on the luminal surface of the intestinal epithelium. This raises cyclic GMP (cGMP), increases secretion of chloride and bicarbonate into the lumen, raises stool fluid content and accelerates transit; pain signaling is also modulated.
Teduglutid
Teduglutide activates the GLP-2 receptor, promoting growth and regeneration of small-intestinal villi, increasing nutrient and fluid absorption and thereby reducing dependence on parenteral nutrition. The glycine substitution at position 2 makes it resistant to rapid breakdown by DPP-4.

Frequently asked questions
- What is the difference between Linaclotid and Teduglutid?
- Linaclotid is classified as "Research other", while Teduglutid is classified as "Research other". Linaclotid: Linaclotide is a 14-amino-acid peptide and guanylate cyclase-C (GC-C) agonist. It is approved for irritable bowel syndrome with constipation (IBS-C) and chronic idiopathic constipation (CIC). Teduglutid: Teduglutide is a DPP-4-resistant analog of GLP-2 (glucagon-like peptide-2). It promotes growth and function of the intestinal mucosa and is approved for treating short bowel syndrome. This page contrasts both neutrally and source-based — with no usage or dosing recommendation.
- Which peptide is better supported by science, Linaclotid or Teduglutid?
- The highest available evidence level is "Human RCT" for Linaclotid and "Human RCT" for Teduglutid. A higher evidence level means more robust data, but says nothing about suitability for an individual. The full body of evidence is on each peptide's own page.
- What is the legal status of Linaclotid and Teduglutid in Germany and the United States?
- Germany: Linaclotid — Prescription, Teduglutid — Prescription. United States: Linaclotid — Prescription, Teduglutid — Prescription. These are factual summaries with source and review date on the individual pages.
